Key to success, is to have an unbiased and open view which synthetic technology is the most suitable. Sometimes this can be a process where a biocatalytic step makes all the difference, while in other cases the safe use of a hazardous intermediate in a flow reactor (flow chemistry) shortcuts the process drastically, or yet in another case a chiral chemocatalyst delivers that specific chiral building block in the most economical way.
A typical example is given below where initially about 6 different routes were shortlisted for a new route to the chiral building block shown in the center. After evaluation we opted successfully for a 2-step route using an phenyl ammonia lyase adding ammonia selectively to the cinnamate, followed by copper catalysed C-N bond formation (all in water).
